Widdrol Induces Apoptosis in Human Cervical Carcinoma HeLa Cells

In this study, the anticancer activity of widdrol, essential oil compound isolated from Juniperus chinensis, is investigated in human cervical cancer HeLa cells. Widdrol inhibits the growth of HeLa cells in a dose- and time-dependent manner. To confirm whether these effects of widdrol are associated with apoptosis, apoptotic features and expression of apoptosis-related proteins are examined. As a result, typical morphological characteristics of apoptotic cells such as formation of apoptotic body was observed as measured by DAPI staining. In Western blot analysis, the expression of anti-apoptotic Bcl-2 and Bcl-XL were decreased and cytosolic cytochrome c was increased in cells treated with widdrol. Also, caspase-8 and caspase-9, which are initiator caspase each in extrinsic and intrinsic pathway, were activated, activation of executioner caspase (caspase-3) and proteolytic cleavage of PARP were also observed. Therefore, widdrol seems to induce apoptosis by activating both intrinsic and extrinsic pathway in HeLa cells at the same time.
